摘要
采用流式细胞术,视频观察和琼脂糖凝胶电泳,观察糖皮质激素曲安奈德预处理对紫杉醇等诱发的人乳癌细胞凋亡反应的影响.曲安奈德预处理可抑制紫杉醇诱导的细胞凋亡,流式细胞术和视频观察显示凋亡抑制率分别为49.3%和52.0%.琼脂糖凝胶电泳显示曲安奈德抑制抗微管剂紫杉醇,秋水仙碱,长春碱所引起的DNA裂解反应,“阶梯状”DNA条带密度降低.对DNA和蛋白质合成抑制剂则无明显影响.甾醇类激素预处理对紫杉醇无明显抑制作用.
To investigate the mechanism by which pretreatment with glucocorticoid alleviates adverse reactions of taxol, flow cytometry, video timelapse monitor and agarose gel electrophoresis were performed to indicate the evidence that pretreatment with triamcinolone acetonide (TA) affects apoptosis of Bcap 37 cells induced by taxol and other drugs. TA pretreatment inhibited taxol induced apoptotic cell death. By flow cytometry and video timelapse monitor, inhibitory rates of apoptotic cells by TA were 49.3% and 52.0%, respectively. Moreover, TA pretreatment also inhibited other antimicrotubule agents(colchicine and vinblastine)induced DNA fragmentation characterized by apoptosis. Pretreatment with steroid sexual hormones had no effect on taxolinduced apoptosis. It is concluded that the pretreatment with glucocorticoid can inhibit taxolinduced apoptosis in human breast cancer cells.
