顺铂对人脐静脉内皮细胞ICAM-1表达的影响

摘要 目的:为细胞间黏附分子(ICAMs)是否参与顺铂引起的血管损伤病理反应寻找证据。方法:将人脐静脉内皮细胞(HUVECs)以不同浓度顺铂(Cisplatin)或同一浓度顺铂处理不同时间后,蛋白印迹分析法和RT-PCR检测内皮细胞CAMs的表达情况,并通过凝胶阻滞分析(EMSA)和特异性抑制剂来探讨NF-κB在其中的作用。结果:顺铂能够在mRNA和蛋白水平明显上调内皮细胞中细胞间黏附分子-1(ICAM-1)的表达,而对P-选择素、E-选择素和VCAM-1的表达无明显影响。其表达是通过核因子κB(NF-κB)依赖途径实现。结论:ICAM-1参与了顺铂的血管毒性病理过程,具有靶向治疗前景。 Objective To explore whether CAMs are involved in the pathogenesis of vascular injury induced with cisplatin. Methods Human umbilical vein endothelial cells (HUVECs) were treated with different concentrations of cisplatin or for different time for cisplatin respectively, the expression of ICAMs in endothelial ceils were analyzed by RT-PCR and Western blot, and the role of NF-κB in the CAMs expression was investigated by EMSA and specific inhibitor. Results Cisplatin could upregulate the expression of ICAM-1 at both mRNA and protein levels ,but had no significant effect on the expression of P-selectin, E-selectin, and VCAM-1, and the expression was through a NF-κB-dependent pathway. Conclusion ICAM-1 involves in the pathogenesis of cisplatin-indueed vascular toxicity and is valuable for clinical targeting therapy.
