摘要 目的:探讨白藜芦醇对宫颈癌Hela细胞生长抑制及诱导凋亡的作用机制。方法:以不同浓度白藜芦醇处理Hela细胞,MTT法测定细胞增殖抑制率;TUNEL法检测细胞凋亡;Western blot检测凋亡相关蛋白Bcl-2、Bax、Bad、Caspase-9的表达。结果:经白藜芦醇处理的Hela细胞,增殖作用呈明显时效和量效关系,细胞凋亡率明显增高并呈浓度依赖性;Bcl-2、Bax、Bad表达下降,Caspase-9表达增加。结论:白藜芦醇对Hela细胞具有增殖抑制和诱导凋亡作用,其机制与下调Bcl-2、Bax、Bad蛋白、上调Caspase-9蛋白表达有关。 Objective To investigate the possible molecular mechanism in resveratrol-induced apoptosis of cervical tumor Hela cell line. Methods Hela ceils were treated with various concentrations of resveratrol, the inhibitory rate of cell prolifer ation was measured by MTT, cell apoptosis was determined by TUNEL assay, and the expression of Bcl-2, Bax, Bad and Caspase-9 protein were detected by Western blotting. Results Resveratrol significantly inhibited the proliferation of Hela ceil line in a time-and dose-dependent manner. The rate of apoptosis was significantly increased and in a dose-dependent manner. The expression of Bcl-2, Bax,Bad was decreasd,but the Caspase-9 expression was increased. Conclusion Resveratrol could inhibit the proliferation and induce apoptosis of human Hela cells. The effects may related to the down-regulation of Bcl-2, Bax, Bad protein expression and up-regulation of Caspase-9 protein expression .
