摘要
以葡萄砧木品种‘5BB’(Vitis berlandieri×V.ripria)为试材,通过设计特异引物、PCR扩增,从‘5BB’基因组中得到1条全长1002bp的多聚半乳糖醛酸酶抑制蛋白(PGIP)基因序歹11,该序列包含1个完整的开放阅读框,与欧洲葡萄、其它果树的相应序列同源性分别为99%和68%,其推导的蛋白质序列中包含一段亮氨酸重复序列,第1~27个氨基酸残基是信号肽,三级结构与菜豆PGIP相似。
Specific primers, which were synthesized according to the conservative regions of PGIP (polygalacturonase inhibiting protein) gene, were used to amplify genomic DNA from grape rootstock '5BB' (Vitis berlandier/ × V. riparia) by polymerase -chain reaction (PCR). A sequence of PGIP gene with whole length of 1002 bp was obtained, which contained a complete open reading frame. Its DNA sequence was homologous to those from Vitis vinifera and other fruits by 99% and 68% respectively. Its protein sequence contained a typical leucine - repeated sequence. The predicted signal peptide was formed by 27 residues from 1 to 27, and the predicted PGIP three dimensional model was similar with that of Phaseolus vulgaris by using bioinformatics method and tool.
