摘要
对水稻细菌性条斑病抗性离体叶鉴定方法的影响因素进行了研究。结果表明:离体叶检验法需在琼脂培养基中加入75~100μL/mL的苯丙咪唑,以保持鉴定叶片非感染部位叶色为绿色;鉴定期间培养温度以25~30℃为宜;接种用茵液浓度不低于3×10^5CFU/mL;当菌液浓度较高时,潜育期的变化对温度不敏感,为5~6d;当菌液浓度与温度同时降低时,则潜育期延长。
The excised leaf tissue inoculation (ELTI) method was used to identify the resistance of rice to bacterial leaf streak in this paper. The results showed that ELTI was a more efficient and dependable method for disease-resistance identification than the traditional method. It was necessary to keep leaf green by adding 75~100μL/mL of benzimidazole into agar medium in the whole identification period. For efficient inoculation and identification of excised leaf tissue, the temperature was required to be 25~30℃, and the lowest concentration of bacterial solution should be 3×10^5 CFU/mL.
