摘要
目的在链脲佐菌素(STZ)糖尿病大鼠模型观察钒酸盐(Va)对骨骼肌和肝脏糖原的影响与cAMP的关系。方法用正钒酸钠分别管饲正常对照大鼠和STZ致糖尿病大鼠,观测肝脏和骨骼肌的糖原与cAMP的变化趋势。结果与正常对照组比较,糖尿病骨骼肌糖原含量下降(P<0.01),肝糖原含量升高(P<0.01);Va使肌糖原上升47%(P<0.01),肝糖原下降50%(P<0.01),均恢复至正常水平,提示至少有部分肝糖原向骨骼肌转移。Va同时升高肝cAMP,降低骨骼肌cAMP,与糖尿病动物使用Va后肌肝糖原变化相反。结论本试验提示cAMP在Va导致的肝糖原向骨骼肌转移中发挥一定作用。
AIM To explore the relationship between changes of glycogen and cAMP induced by vanadate (Va). METHODS The contents of cAMP and glycogen were measured and their changes observed in liver and skeletal muscle isolated from normal control rats and streptozotocininduced diabetic rats, some of which were gavaged with sodium orthovanadate solution, others with physiological salt solution as controls. RESULTS ① Glycogen content decreased (P<001) and increased (P<001), respectively, in diabetic skeletal muscle and liver when compared to normal rats; after Va treatment (2 weeks), the former elevated by 47% (P<001), the latter reduced by 50% (P<001), both near to normal level. This suggested at least part of liver glycogen was transferred to skeletal muscle via blood circulation by Va; ② The change of cAMP in skeletal muscle and liver is opposite to that of glycogen in diabetic rat after Va treatment. CONCLUSION cAMP plays certain role in the mechanism of glycogen transfer from liver to skeletal muscle induced by Va.
