摘要
目的观察大黄素对角朊细胞体外增殖分化的影响。方法以人转化型角朊细胞株COLO-16细胞为模型,采用活细胞计数、3H-TdR参入法、细胞因子生物活性检测、电镜及ABC法进行研究。结果大黄素能以一种剂量依赖方式抑制角朊细胞的体外增殖和DNA合成;减少角朊细胞肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)的产生;对角朊细胞超微结构有直接损伤作用;并抑制角朊细胞角蛋白表达,破坏细胞骨架。结论大黄素通过不同途径抑制角朊细胞的体外增殖分化,是角朊细胞增殖分化的有效抑制剂。
AIM The effects of emodin on keratinocyte proliferation and differentiation in vitro were observed. METHODS Using the culture of human transformed keratinocyte cell line COLO16 cells as a model, viable cell count, 3HTdR uptake, cytokine bioactivity assay, electron microscope examination and ABC method were applied in this study. RESULTS The inhibitory effects of emodin on keratinocyte proliferation and DNA synthesis were observed, and it was found that the inhibitory effects were dosedependent. In the supernatant dirived from COLO16 cells treated with emodin, tumor necrosis factorα (TNFα) and interleukin6 (IL6) production were significantly reduced. The membrane, mitochondrion and cytoskeleton of keratinocytes were severely damaged and the keratin expression was suppressed. CONCLUSION This study suggestes that the proliferation and differentiation of keratinocytes in vitro were suppressed by emodin through various ways, and emodin may be an effective suppressing drug on keratinocyte proliferation and differentiation.
