优化喜树果中喜树碱和10-羟基喜树碱的匀浆提取工艺 被引量:4

Optimization of Homogenate Extraction of Camptothecin and 10-hydroxycamptothecin in Seeds of Fructus camptothecae acuminatae

摘要 采用匀浆提取的方法从喜树果中提取喜树碱和10-羟基喜树碱,考察乙醇体积分数、匀浆时间和液料比等因素对提取率和纯度的影响,并通过响应面法进行工艺优化,分别得到以喜树碱、10-羟基喜树碱的提取率和纯度为响应值的回归方程,进而获得最佳工艺条件为乙醇体积分数52.41%;匀浆时间2.93 min;液料比11.11(mL∶g)。最佳条件下的验证实验表明:喜树碱提取率为0.092 3%,浸膏中喜树碱的纯度为1.063 3%,与模型值相对偏差分别为0.75%和0.88%;10-羟基喜树碱提取率为0.009 4%,浸膏中10-羟基喜树碱的纯度为0.107 8%,与模型值相对偏差分别为0.53%和0.78%,说明回归方程可以很好地预测实验结果。 Camptothecin (CPT) and 10-hydrxycamptothecin (HCPT) in seeds of Fructus camptothecae acuminatae were extracted by homogenate method. The effects of volume fraction of ethanol, homogenate time and ratio of liquor to material on the extraction yield and purity were investigated and the process was optimized by response surface methodology (RSM). The regression equations based on the extraction yield and purity of CPT and HCPT were obtained, further, the optimum technology parameters were obtained and they were that 52.41% of ethanol volume fraction, 2.93 min of homogenate time and 11.11 (mL: g) of liquor to material. Under the optimum condition, the extraction yield of CPT was 0.092 3 %, the purity of CPT in extractum was 1.063 3%, and the relative deviations of experimental and simulation values were 0.75% and 0.88%, respectively. The extraction yield of HCPT was 0.009 4% and the purity of HCPT in extractum was 0.107 8%. The relative deviations of experimental and simulation values were respectively 0. 53 % and 0. 78 %, which showed regression equations can well predict the experimental results.
