摘要
目的:探讨EZH2基因在增生子宫内膜、非典型增生子宫内膜、子宫内膜癌中的表达及其与子宫内膜癌发生、发展及预后的关系。方法:采用RT-PCR方法检测21例增生子宫内膜、27例子宫内膜不典型增生及46例子宫内膜癌组织中EZH2 mRNA的表达,并对结果进行统计学分析。结果:EZH2 mRNA在增生子宫内膜、不典型增生子宫内膜、子宫内膜癌组织中的表达分别为0.326±0.163、0.548±0.291、0.817±0.126,呈逐渐增高趋势,统计学分析显示各组间差异有统计学意义(P<0.05);子宫内膜癌组中EZH2表达水平与临床各病理参数密切相关,随着组织分化程度降低、肌层浸润加深和手术-病理分期增高EZH2表达逐渐增加(P<0.05);与淋巴结转移无关(P>0.05)。结论:EZH2在子宫内膜腺癌的发生、发展过程中起一定作用,EZH2的过表达促进了肿瘤的发生,它的检测对子宫内膜腺癌的早期诊断、恶性程度的判定有积极意义。
Objective: To explore the expression of enhancer of zeste homolog 2 ( EZH2 ) gene in proliferative endometrium, atypical hyperplasia endometrium, endometrial carcinoma and the relationship between EZH2 expression and oncogenesis, progression and prognosis of endometrial carcinoma. Methods: The expression levels of EZH2 gene in 21 cases of proliferative endometrium, 27 cases of atypical hyperplasia endometrium and 46 cases of endometrial carcinoma were detected by real - time fluorescent quantitative reverse transcription - polymerase chain reaction ( RT - PCR) . Results: There was significant difference in the expression of EZH2 mRNA between proliferative endometrium (0. 326 ±0. 163), atypical hyperplasia endometrium (0. 548 ±0. 291 ) and endometrial carcinoma (0. 817 ±0. 126) (P 〈 0.05 ) . The expression of EZH2 was related to pathologic parameters, the expression of EZH2 increased with low histodifferentiation, deep muscular layer invasion and senior surgical - histological stages ( P 〈 0. 05 ) , while the expression of EZH2 was independent of lymphatic metastasis ( P 〉 0. 05 ) . Conclusion : EZH2 maybe play an important role in the oncogenesis and progression of endometrial adenocarcinoma. The high level of EZH2 promotes the oncogenesis of endometrial adenocarcinoma. Detection of EZH2 gene has positive effects on early diagnosis and judgement of malignant degree in endometrial carcinoma.
