肺癌组织中抗氧化酶及氧化代谢物变化的定量分析

摘要 背景与目的抗氧化酶与肿瘤的关系逐渐受到重视。通过对113例原发性肺癌患者手术切除肿瘤组织和肿瘤远端正常肺组织的抗氧化酶和氧化代谢物及SODmRNA及其蛋白表达量的测定,观察肺癌组织氧化/抗氧化状态的特点,为补充或完善肺癌发生和发展机制提供一定的依据。方法用比色法测定肺癌组织抗氧化酶活性和氧化代谢物含量,包括:总超氧化物歧化酶(TSOD)(包括MnSOD和CuZnSOD)、谷胱甘肽过氧化物酶(GPx)、过氧化氢(H2O2)、丙二醛(MDA),用半定量PCR和Westernblot法测定患者组织中MnSOD和CuZnSOD的mRNA和蛋白表达量。结果①与肿瘤远端正常肺组织相比,肺癌组织中的TSOD、CuZnSOD、GPx活性增高,H2O2、MDA降低(P〈0.05或0.01)。②肺腺癌组织中的氧化代谢物H2O2、MDA高于肺鳞癌组织(P〈0.05或0.01)。③与肿瘤远端正常肺组织相比,肺癌组织中MnSODmRNA和蛋白表达量降低,CuZnSODmRNA和蛋白表达量增高(P〈0.01)。结论肺癌组织存在着氧化/抗氧化失衡。SOD的异常表达水平均出现细胞异常生长,提示氧化/抗氧化失衡与肺癌的发生和发展存在一定的关系。 Background and Objective The relationship between antioxidants and tumor was gradually regarded.The changes of oxidants and antioxidants in the activities,the expression of mRNA and protein were evaluated in 113 lung tumor and 64 tumor-free lung tissues collected between the years 2006 and 2007 from 113 individuals with surgically resectable lung cancer.To consummate the mechanism of the onset and development of lung cancer.Methods The activities of TSOD,MnSOD,CuZnSOD,GPx,H202 and MDA were determined by chromometry. MnSOD,CuZnSOD mRNA and protein expression were evaluated by reverse transcription polymerase chain reaction and Western blot analysis in tumor,as well as corresponding control tissues. Resulst ①To compare with the level of these indexes in the tumor-free tissues,the activities of TSOD, CuZnSOD, GPx were higher, H2O2 and MDA were lower in the tumor tissues(P〈0.05 or 0.01).②To compare with the group of the squamous cell carcinoma,H2O2,MDA were higher in the tumor tissues of the group of adenocarcinoma(P〈0.05 or 0.01). ③There was less MnSOD mRNA and protein in tumor than in tumor-free lung.There was more CuZnSOD mRNA and protein in tumor than in tumor-free lung tissue(P〈 0.01). Conclusion The imbalance between the oxidants and antioxidants exists in the tissues of patients with lung cancer.The imbalance may have some relationship with the onset and development of lung cancer.
