复方黄金香提取液对创伤性脑水肿大鼠AQP4表达的影响初探

摘要 目的通过观察复方黄金香提取液对水通蛋白4(aquaporin4,AQP4)表达的影响,探讨其治疗创伤性脑水肿的相关机制。方法SD大鼠40只,参照Feeney法制作脑损伤模型(TBE模型),随机分组:祛瘀通腑汤组大鼠(10只)致伤后20 min予以祛瘀通腑汤10 mL.kg-1灌胃,复方黄金香提取液组大鼠(10只)致伤后20 min予以复方黄金香提取液10 mL.kg-1灌胃,对照组(10只)和创伤组(10只)同时同法给予相同剂量生理盐水。按12、24、72 h分别处死大鼠,取标本分析AQP4表达的影响,并作HE染色。结果"复方黄金香提取液"与"祛瘀通腑汤"相比明显减轻脑水肿,其AQP4表达水平显著下调。病理学证实,"复方黄金香提取液"使脑组织水肿明显减轻,神经元坏死数目明显较少。结论早期应用"复方黄金香提取液"可明显减轻脑水肿程度;下调AQP4表达是其作用机制之一。 Objective To determine the mechanism of compound Huangjinxiang extraction liquid (HJEL) for traumatic brain edema (TBE) by observing changes of auuaporin 4 (AQP4) expression in the brain tissues. Methods According to Feeney methods (TBE), 40 SD rats were randomly divided into a control group (10 rats), a trauma group (10 rats), a Quyutong futang (ESC) group (10 rats), and an HJEL group (10 rats), the rats in Quyutongfutang group were intragastrically administered ESC at 10 mL·kg^-1 while the rats in the HJEL group were intragastrically administered HJEL at 10 mL·kg^-1. The rats in the trauma group, the ESC and HJEL group were injured with Feeney injury models. The rats were euthanized for bistopathologic studies and for AQP4 expression. Results AQP4 expression was significantly lower in the HJEL group than in the ESC group. Histopathologic studies showed more surviving neurons and less edema in the brain tissues in the HJEL group than in the ESC group. Conclusion Early administration of HJEL can alleviate brain edema effectively, and downregulation of AQP4 expression is part of its mechanism.
