人类衰老性疾病与Klotho基因的密切关系

摘要 Klotho基因是与人类衰老密切相关的基因,该基因表达产物有膜结合型和分泌型2种蛋白异构体,二者分别是体液调节信号蛋白和膜结合受体。klotho基因缺陷鼠可出现一系列与人类衰老相似的多种行为和病理变化:如寿命缩短、性功能障碍、痴呆、白内障、运动失调、肺气肿、动脉粥样硬化、骨质疏松以及免疫功能降低等。使用一定的方法使小鼠重新获得基因,各种症状均能缓解。研究者认为klotho基因敲除鼠是目前研究人类衰老及多种老年病发生、预防和治疗的最合适的动物模型。
