家蚕表达的重组人巨噬细胞集落刺激因子抗病毒活性的体外研究

摘要 以家蚕表达的重组人巨噬细胞集落刺激因子(rhMCSF)预先处理细胞,再接种不同病毒,观察rhMCSF对病毒增殖的影响。结果显示,rhMCSF在人胚皮肌细胞上对所观察的单纯疱疹病毒1和2型、流感病毒A3型、鼻病毒、水泡性口炎病毒、腺病毒6和11型均有不同程度的抑制作用,推迟或减轻细胞病变的发生;在单纯疱疹病毒1型感染乳兔肾细胞的实验中能使子代病毒滴度降低100倍以上,TNFα、IFNα和γ的中和单抗均不能阻断这一作用,而TNFβ的中和单抗却能部分阻断这一作用。 Pretreated with recombinant human macrophage colony stimulating factor(rhM CSF) expressed in silkworm,the cells were inoculated with several viruses and then effect of the rhM CSF on viral replication was observed.The results showed that in the cultures of fibroblast derived from human fetal skin muscle tissues infected with the viruses observed in this experiment,including HSV 1 and 2,influenza virus A3,rhinovirus,VSV,adenovirus 6 and 11,rhM CSF inhibited the virus induced cytopathic effect to different degrees,defering or relieving the cytopathy and reduced the titer of HSV 1 in the infected cells derived from breast rabbit kidney over 100 fold.This antiviral activity could be neutralized by anti IFN β McAb,not by anti IFN α,anti IFN γ and anti TNF α McAbs,indicating the relationship of the mechanism of anti virus activity of rhM CSF to the induction of IFN β.
