摘要
目的:探讨大鼠耳蜗是否存在有内源性ANP,并试图定位耳蜗的ANP免疫反应细胞和阐明调节迷路液体衡定的机制。方法:RAN斑点杂交技术;PAP法免疫组织化学技术。结果:斑点杂交结果:蜗轴总RNA斑点杂交呈浅紫色阳性反应。免疫组织化学观察到蜗管的ANP免疫反应细胞分布于:蜗管外侧壁的外沟细胞的胞浆内显示出很强的棕色反应产物。外沟细胞的"根"延伸至螺旋韧带的结缔组织内。螺旋韧带组织呈弱棕色反应,但在螺旋韧带中、上部比其下部的反应产物颜色稍深。大多数螺旋神经节细胞为ANP免疫反应阳性细胞,棕色ANP免疫反应产物散布在胞浆内,少数神经节的胞核也被阳性染色。在螺旋神经节内的神经纤维及蜗轴内的蜗神经都有ANP免疫反应阳性纤维。结论:内源性ANP是由下列两种细胞产生:即蜗管外侧壁的外沟细胞和蜗轴的螺旋神经节神经元。外沟细胞分泌的ANP可通过它的"根"和螺旋韧带输送到血管纹和螺旋隆凸,影响内淋巴离子成分的形成,参与调节迷路液体的平衡,这可能是蜗管内的一种局部调节。
Objective:To approach whether there has been endogenous ANP in the rat cochlea,and also to try positioning the immunological reaction cell of the rat cochlea,and to interpret the regulation mechanism of the constancy of labyrinthine fluid.Methods: RAN dot blot hybridization;PAP immunohistochemistry.Results:The immunological reaction cell of the rat cochlea disposited in the lateral wall,the intracytoplasm of ecto-canal cell displayed brown response entity by immunohistochemistry.The “root” of the ecto-canal cell extended to the spiral ligament.The spiral ligament displayed feeble brown reaction,nevertheless in the middle part and superior part the response entity displayed deep color compared with the subjacent part.The majority spiral ganglion cell displayed positive in ANP immunological reaction,brown ANP immunological reaction scattered in the intracytoplasm,and minority ganglionic nucleus displayed positive staining.There has been positive fiber in the neurofibra of spiral ganglion and in the cochlea nerve of modiolus by ANP immunological reaction.Conclusion: Endogenous ANP generated from two cells:the ecto-canal cell in the lateral wall of cochlea duct,and the ganglionic neuron of spiral ganglionic in the modiolus.ANP secreted by ecto-canal cell has been transported to psalterial cord and spiral prominence by the “root” and spiral ligament,thereby to affect the formation of the endolymph ion and to participate regulate the constancy of labyrinthine fluid.Maybe that is a local regulation.
