摘要
目的:建立HPLC测定盐酸哌甲酯血药浓度的方法。方法:血浆样品经碱化后用乙醚提取,采用高效液相色谱-荧光检测法(HPLC-RF)测定血浆中盐酸哌甲酯的浓度,采用ODS C18(4.6mm×150mm,5μm)色谱柱,流动相为乙腈-0.03 m·L^(-1)磷酸缓冲液(22:78);荧光检测激发波长为252nm,发射波长为295nm。结果:血浆哌甲酯浓度在(2~100)μg·mL^(-1)范围与其峰高比呈良好的线形关系,r =0. 999;平均加样回收率为102.5% , RSD为6.0%。结论:本实验方法灵敏、准确、简便,适用于人血浆样品的哌甲酯浓度测定。
To establish a simple and specific high-performance liquid chromatographic (HPLC) method to determine the concentrations of methylphenidate hydrochloride in Human Plasma.Methods:Chromatography was performed with ODS C18 column (4.6mm×150mm,5μm) and a mobile phase of acetonitrile-0.03 mol·L^-1phosphate buffer (22:78),with RF detection at Ex 252 nm and Em 295nm.Results:The calibration curve was linear range of (2-100)μg·mL^-1 (r=0.999).The average extraction recovery of methylphenidate hydrochloride in human plasma was 102.5%,and RSD was 6.0%.Conclusion:The analytical method was found to be applicable,reliable and suitable for studies.
