摘要
目的:对昆布多糖进行硫酸酯化,并对产物的纯度与结构进行分析,为研究其生物活性奠定物质基础。方法:利用氯磺酸-吡啶法对昆布多糖进行硫酸酯化,并使用HPLC-ELSD法测定反应产物的纯度与分子量;采用硫酸钡比浊法测定昆布多糖硫酸酯中SO42-的含量,并利用波谱法研究其结构。结果:昆布多糖硫酸酯的纯度为96%,其分子量为16315,SO42-含量为41.30%;紫外、红外光谱分析结果均显示硫酸酯化多糖的光谱特征,13C-NMR谱显示多糖的2位羟基为SO42-的主要取代位置。结论:利用氯磺酸-吡啶法对昆布多糖进行硫酸酯化,该方法所得产物的纯度和酯化率高。
Objectives: To sulfate the laminarin, then analyze the purity and elucidate the structure of polysaccharide sulfate, so as to make a foundation for the further research about its biological activity. Methods: LAMS was prepared by chlorsulfonie acid-pyridine method, and the purity and molecular weight was analyzed by HPLC - ELSD. The content of SO4^2- in polysaccharide sulfate was determined by barium sulfate turbidimetry, then the structure of polysaccharide sulfate was elucidated by UV, IR and 13C -NMR. Results: The purity of LAMS was 96%, the molecular weight was 16315 and the contents of SO4^2- was 41.30%. Both UV and IR spectrum showed the characteristic of polysaeeharide sulfate, and the 13 C - NMR exhibited that most of the SO4^2- were on 2 - OH. Conclusion: LAMS was prepared from laminarin by chlorsulfonic acid-pyridine method. The rate of esterification and the purity of production are high.
