摘要
目的观察聚乳酸—聚乙醇酸共聚物(PLGA)-Ⅰ型胶原—壳聚糖复合人工硬脊膜的生物相容性。方法制作PLGA膜(膜Ⅰ)、PLGA-Ⅰ型胶原复合膜(膜Ⅱ)、PLGA-Ⅰ型胶原—壳聚糖(9∶1)复合膜(膜ⅢA)、PL-GA-Ⅰ型胶原—壳聚糖(5∶5)复合膜(膜ⅢB),对其行接触角、吸水率测定及细胞毒性实验。结果吸水率:膜Ⅰ<膜ⅢB<膜ⅢA<膜Ⅱ,P均<0.01;接触角:膜Ⅱ<膜ⅢA<膜ⅢB<膜Ⅰ,P均<0.01;细胞毒性实验:第1天,各膜间OD值比较,P>0.05;第3、7天,膜Ⅰ与膜Ⅱ、膜ⅢA,膜ⅢA与膜ⅢB比较,P均<0.05。结论PLGA膜经Ⅰ型胶原和壳聚糖改性后,可以促进细胞在膜上的黏附、贴壁能力。膜ⅢA在生物相容性方面基本符合人工硬脊膜的要求。
Objective To observe the biocompatibility of PLGA-type- Ⅰ eollagen-Chitosan composite membrane as arti- ficial spinal dura mater. Methods To produce PLGA membrane ( M Ⅰ ) , PLGA-type- Ⅰ collagen composite membrane (MⅡ) ,PLGA-type- Ⅰ collagen-Chitosan(9:1 ) composite m (M Ⅲ A) and PLGA/-type- Ⅰ collagen-Chitosan(5:5 ) composite (M ⅢB). Contact angle, absorption rate and eytotoxieity study were used to research all type ms. Results Contact angle: M Ⅱ 〈 MⅢA 〈 M B 〈M Ⅰ ,P 〈0.01 ;Absorption rate:MⅠ 〈 MⅢB 〈 MⅢA 〈 M Ⅱ ,P 〈 0.01 ; Cytotoxic experiment:at 1 st day, the OD value between each membrane didn' t have significant difference, P 〉 0.05. At 3rd and 7th day, there is significant difference between M Ⅰ and Ⅱ or mA, ( M ⅢA and ⅢB, all P 〈 0.05. Conclusions After being modified by type- Ⅰ collagen and Chitosan, PLGA membrane could striking enhance the adhesion and proliferation of L929 cell. On the whole, ( M ⅢA can meet the biocompatibilitv as a tvpe of material of artificial sninal dura mater.
