摘要
目的采用体外实验研究脆弱类杆菌脂多糖(LPS)对表皮细胞的损伤作用,探讨脆弱类杆菌的致病机制。方法用不同剂量的脆弱类杆菌脂多糖(LPS)刺激体外培养的人表皮细胞,24h后测定上清液乳酸脱氢酶(LDH)浓度及细胞增殖活力,并进行相关性分析。结果与空白对照组比较,脆弱类杆菌LPS刺激后培养上清液中LDH含量未见显著性增加(P>0.05),对细胞增殖活力也无明显抑制作用。而阳性对照组大肠杆菌LPS刺激后上清液中LDH含量显著增加,细胞活力显著下降。结论脆弱类杆菌脂多糖对体外培养人体表皮细胞无明显损伤作用,脆弱类杆菌LPS在脆弱类杆菌引起的炎症反应中并非重要的致病物质。
Objective To explore the virulent mechanism of lipopolysaccharide coming from bacteroides fragilis on cultured human ecpidermal cell and it' s function in the infection. Methods Cultured human eepidermal cell and added lipopolysaccharide into culture medium, studied the changes of LDH quantity in medium and cells' proliferation. Their correlations were analyzed. Results The LDH quantity in medium increased and the cells' proliferation slowed after cells induced by LPS from coliform, but they are not changed after induced by LPS from bacteroides fragilis. Condusion lipopolysaccharide of bacteroides fragilis can' t inhibit ecpidermal cell alone. The LPS is not an important pathogenic factor of bactroides fragilis.
