摘要
目的研究急性髓性白血病(Acute myeloid leukaemia,AML)病人的多药耐药基因(Multidrugresistance)MDR-1G2677T的多态性和P-gp(Permeability-glycoprotein)蛋白表达的相关性。方法采用流式细胞仪技术测定P-gp表达水平,限制性内切酶多型性-聚合酶链反应测定MDR-1G2677T多态性,并统计其相关性。结果中国AML病人中MDR-1G2677T基因座上的多态性有自己的分布特点,MDR-12677GG倾向于表达较高的P-gp。结论黄种人AML病人中MDR-1G2677T基因座上的多态性有与白种人不同的分布特点,可能为不同种族的AML病人对相同的化疗药物有效率却不同的原因之一;MDR-12677GG预测AML病人化疗敏感程度较其它携带者低,可以作为不良预后因素之一。
Objective To evaluate the correlation of MDR - 1 G2677T polymorphism and P - gp expression in AML peripheral blood sample. Methods P - gp expression was measured by flow - cytometry technology, while MDR - 1 G2677T polymorphism was analyzed by RFLR - PCR. The correlation was examined by Kruskal - Wallis tes, Results blDR - l G2677T polymorphism in Chinese AML patients has its unique distribution which is disparate from Caucasian. Patients carried MDR - 1 2677GG tends to have high P - gp expression. Conclusion The various distributions of MDR - 1 polymorphisms maybe contribute to different response rate in different races AML patients. MDR - 1 2677GG alleles tends to correspond to high P - gp expression which possibly predicts a un- favorable outcome in AML patients.
