摘要
目的研究FlexRate法在雅培AEROSET2000全自动生化分析仪上检测超高酶活性样本的应用价值。方法应用FlexRate法测定丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)、碱性磷酸酶(ALP)、γ-谷氨酰转移酶(GGT)、肌酸激酶(CK)和淀粉酶(AMY),对FlexRate法的精密度和线性范围进行测定,并将该法测定与稀释模式测定的结果进行对比。结果FlexRate法测定ALT、AST、ALP、GGT、CK、AMY的检测上限分别提高到3 356、3 194、4 368、2 955、5 437、4 878 U/L,该法测定6种酶的批内变异系数(CV)均小于常规法。FlexRate法测定与稀释模式测定的结果比较,相关性良好。结论应用FlexRate法测定6种酶活性,重复性良好,提高了检测上限,FlexRate法测定与稀释模式测定的结果有良好的相关性。FlexRate法测定可大大减少稀释重做的次数,提高工作效率。
Objective To investigate the application value of FlexRate method for detecting enzyme activities in the Abbott AEROSET 2000 automatic analyzer. Methods The precision and linearity of FlexRate method used to detect alanine aminotransferase ( ALT), aspartate aminotransferase (AST) , alkaline phosphatase (ALP),γ-glutamyhransferase (GGT), creatine kinase(CK), and amylase (AMY) were determined. The FlexRate results were compared with the results obtained by re - analysis with dilution model. Results The upper limits of FlexRate method for detecting ALT, AST, ALP, GGT, CK,and AMY were extended to 3 356 U/L,3 194 U/L,4 368 U/L,2 955 U/L,5 437 U/L,and 4 878 U/L, respectively. The within - run coefficients of variation(CV) in FlexRate method were lower than those in routine method. The results obtained by FlexRate and re - analysis with dilution model were in good agreement. Conclusions The FlexRate method with good pre- cision extends the linearity of six enzyme assays. The correlation of FlexRate results with values obtained by dilution model are in good agreement. The re -analysis frequency with dilution model is reduced and laboratory efficiency is enhanced by using FlexRate.
