高效液相色谱检测转基因生菜中白藜芦醇和白藜芦醇苷的研究 被引量:2

Determination of Resveratrol and Piceid in Transgenic Lettuce by High Performance Liquid Chromatography

摘要 目的建立快速检测转基因生菜中白藜芦醇和白藜芦醇苷的高效液相色谱法。方法采用Agilent-ZORBAX SB-C185μm4.6×250mm色谱柱,流动相为乙腈-水(45+55),流速1.0ml/min,波长303nm。结果线性关系良好,白藜芦醇r=0.9999,回收率86.0%~97.8%,变异系数6.6%,最低检出量0.0113μg;白藜芦醇苷r=0.9991,回收率81.6%~95.6%,变异系数7.0%,最低检出量0.0159μg。结论以高效液相色谱检测转基因生菜中白藜芦醇和白藜芦醇苷,操作方便、快速、准确度高。 Objective A method was established to determine the content of resveratrol and piceid in transgenic lettuce by high performance liquid chromatography (HPLC). Methods A chromato bar (Agilent ZORBAXSB C18 5μm 4.6 ×250mm) was used with 1.0 ml /min acetonitrile water as mobile phase and detection wave length was 303 nm. Results Good linearities of the calibration curves of resveratrol (r = 0. 9999) and piceid (r 0. 9991 ) were obtained. The recovery rate of resveratrol was 85.0- 97.8%, the variation coefficient was 6.6%, and the detectable limit was 0.0113μg. The recovery rate of piceid was 81.6-95.6 %, the variation coefficient was 7.0 %, and the detectable limit was 0.0159 μg. Conclusions The method is simple, rapid and accurate, and is suitable to determine resveratrolin and piceid in transgenic lettuce.
