摘要
目的:评价风湿病自身抗体蛋白芯片的临床应用价值。方法:用风湿病自身抗体蛋白芯片对375份临床标本进行类风湿因子(RF)、抗环瓜氨酸肽(CCP)抗体、抗角蛋白抗体(AKA)、抗核周因子(APF)、抗核抗体(ANA)的检测,并与上述指标各自经典的检测方法结果进行对比。结果:风湿病自身抗体蛋白芯片法与免疫比浊法对RF、ELISA法对CCP和免疫荧光法对AKA、APF及ANA的检测中均具有较好的一致性(P>0.05)。其中RF与免疫荧光法检测结果阳性符合率为94.7%,阴性符合率为96.1%;CCP与ELISA法检测结果阳性符合率为87.5%,阴性符合率为96.2%;AKA与免疫荧光法检测结果阳性符合率为95.5%,阴性符合率为99.3%;与免疫荧光法检测结果阳性符合率为94.7%,阴性符合率为96.1%;ANA与免疫荧光法检测结果阳性符合率为91.5%,阴性符合率为99.3%。结论:风湿病自身抗体蛋白芯片试剂盒具有操作简便、快速和特异性强等优点,能有效的提高对类风湿疾病诊断的准确率,值得在临床上推广使用。
Objective:To evaluate the clinical value of Rheumatism auto-antibodies using the protein chip method. Methods: A protein chip method were employed to detect rheumatoid factor( RF), anti-cyclic citrullinatod peptide (CCP) antibody, anti-keratin antibody (AKA) , anti-perinucliar factor (APF) and anti-nuclear Antibody (ANA) of 375 clinical specimens, and the results were compared with its classical laboratory methods respectively. Results: The detect ing results of RF, CCP; AKA, APF and ANA with protein chip are consistent with their respective classical results ( P 〉 0. 05 ). In RF the coincidence rate of positive results of protein chip and Immunoturbidimetry is 94.7%, the coincidence rate of negative result is 96. 1% In AKA, APF, and ANA the coincidence rates of positive results of protein chip and Immunofluorescence are respectively 95.5 % ,94. 7 % and 91.5 %, while the coincidence rate of negative results are respectively 99. 3 %, 96. 1% and 99.3% ;In CCP, the coincidence rate of positive results of protein chip and ELISA iS 87.5%, the coincidence rate of negative result is 96.2%. Conclusions : Rheumatic auto-antibodies detected by protein chip method is simple, rapid and specific,which can effectively improve the diagnositic accuracy of rheumatoid diseases, and it is worth of clinic application.
