高压力及曲格列酮对THP-1源泡沫细胞ATP结合盒转运子A1表达的影响

Effect of high hydrostatic pressure and Troglitazone on ATP binding cassette transporter A1 expression in TPH-1 macrophages-derived foam cells

摘要 目的观察高压力及曲格列酮干预对THP-1源泡沫细胞ABCA1 mRNA及蛋白表达的影响。方法将THP-1源泡沫细胞分为9组:对照组、120组、140组、160组、180组、120T组、140T组、160T组和180T组。对照组、120组、140组、160组和180组分别在0~180mmHg压力下培养48h;其余4组则先给予曲格列酮干预后,再分别在相应压力下继续培养48h,终止实验。结果120组、140组、160组和180组ABCA1 mRNA表达较对照组明显减少,120T组、140T组、160T组和180T组与相应组比较,mRNA表达明显增加,差异有显著性(P<0.05或P<0.01),120T组和140T组mRNA表达与对照组相当(P>0.05),ABCA1蛋白表达与mRNA类似。结论高压力下调了THP-1源泡沫细胞ABCA1 mRNA和蛋白表达。曲格列酮能阻断或减弱这种作用,提示高压力减少ABCA1 mRNA和蛋白表达的作用与PPARγ有关。 [Objective] To investigate the effects of different high hydrostatic pressure and troglitazone on the expression of ABCA I (mRNA, protein) in THP-1 macrophage -derived foam cells. [Methods] THP-1 macrophage - derived foam cells were divided into nine groups: control group, 120 group, 140 group, 160 group ,80 group which were incubated under different hydrostatic pressure from 0 to 180 mmHg for 48 hours, and 120T group, 140T group, 160T group, 180T group which were incubated in the same way after being pretreated with 10.0 μmol/L troglitazone for 24 hours. The changes of ABCA 1 mRNA and protein were determined by reverse trancriptase polymerase chain reaction (RT-PCR) and Western blot. [Results] The expression level of ABCA I mRNA and protein in THP-I macrophage -derived foam cells were significantly lower (P 〈0.05 or 0.01) in 120 group, 140 group, 160 group and 180 group comparing with that of control group, and they were significantly higher in 120T group, 140T group, 160T group and 180T group comparing with that in the same pressure group respectively. [ Conclusion] From 120 mmHg to 180 mmHg , high pressure decreased markedly the expression of ABCA 1 (mRNA, protein) in THP-1 macrophage -derived foam cells, which could be markedly blocked or weakened by Troglitazone.
