摘要
选择滤纸作基质,以LiAc作重原子微扰剂,首次成功地建立了测定痕量中草药有效成分葛根素的滤纸基质室温燐光法。本法取样量少(2μL),线性范围宽(4.16~499ng/斑),灵敏度高(检测限为0.19ng/斑),操作简便、快速。
A solid surface room temperature phosphorimetry of puerarin was developed. The maximal excitation wavelength is at 348 um and emission wavelength is at 455 um with a shoulder at 472 um. Several solid substrate and heavy-atom perturbers were tested, filter paper and LiAc were chosen as the substrate and the heavy-atom perturber respectively for the experiment. Other factors influencing the room temperature phosphorimetric intensity such as drying temperature, drying time and pH, were also studied. The linear dynamic range is 4.16-499 ng, the detection limit is 1.17 ng/spot.
