摘要
目的:建立测定蛹虫草子实体正丁醇部位腺苷含量的高效液相色谱(HPLC)分析方法,并在此基础上初步建立正丁醇部位的特征图谱。方法:利用Angilent TC-C18(250mm×4.6mm,5μm)色谱柱,甲醇5%~30%为流动相梯度洗脱,分析时间60min,检测波长260nm,流速1.0mL·min-1,柱温25℃;对样品提取条件,萃取条件,流动相,检测波长等进行了系统优化设计。结果:测定了11个批次的蛹虫草正丁醇部位样品腺苷的含量平均值为0.536%,初步构建了蛹虫草子实体正丁醇部位HPLC特征图谱。结论:该方法准确简便,重复性好,可作为控制蛹虫草子实体正丁醇提取物内在的质量标准。
OBJECTIVE To establish an HPLC method for the assay of adenosine in n-butyl alcohol effective extract of cordyceps Militaris as well as its specific chromatogram. METHODS The determination were carried out by HPLC under the following condition: Angilent TC-C18 column(250 mm×4. 6mm, 5 μm), methanol 5%-30% in 60 min as elution, detective wavelength at 260 nm, flow rate of 1.0 mL·min^-1 and column temperature of 25 ℃ ; The systematic optimized design of the chromatographic conditions, such as supersonic extraction, mobile phase and detective wavelength as well was carried out. RE- SULTS The average proportion of adenosine in n-butyl alcohol effective extract of 11 batches of samples is 85.36%. Based on this method, the HPLC fingerprint of its effective extract was established. CONCLUSION The method is simple, accurate, re-producible and useful for the quality control of cordyceps militaris.
