摘要
目的研究丹参酮ⅡA(TanⅡA)对人卵巢癌A2780细胞增殖抑制及诱导凋亡的作用。方法以四甲基偶氮唑蓝(M TT)法测定TanⅡA对人卵巢癌A2780细胞的增殖抑制;流式细胞仪检测细胞周期。结果TanⅡA对人卵巢癌A2780细胞的生长具有抑制作用,其作用强度随药物作用时间及浓度的增加而增强。实验组较对照组G0/G1期细胞数量增多,而S期细胞数量减少。4.0mg/L TanⅡA作用0、24、48、72h后的凋亡率分别为4.52%、19.7%、25.3%、40.4%,随时间的延长而凋亡细胞增多。结论TanⅡA对人卵巢癌A2780细胞有明显的增殖抑制和诱导凋亡的作用,且其作用呈时间-剂量依赖性。
Objective: To observe the effects of inhibition and inducing apoptosis of tanshinone ⅡA on human ovarian cancer A2780 cell lines. Methods: MTr assay was used to analysis the inhibitive action of tanshinone ⅡA to the A2780 cell line. The cell cycle was determined by flow cytometry. Results: Tanshinone ⅡA could inhibit human ovarian cancer A2780 proliferation, and the intensity increases with time and the concentration of drug. The experimental group had much more G0/G1 phase cells and less S phase cells than the control group. The A2780 cell lines treated by 4.0 mg/L tanshinone HA for 0, 24, 48 and 72 h,and the apoptosis rates were 4. 52%, 19.7%, 25.3%, 40.4%. It showed a time - dependence. Conclusion: Tanshinone ⅡA can significantly inhibit the proliferation of human ovarian cancer A2780 cells and induced the cells apoptosis, and it show a time - dose - dependence.
