摘要
考察了所制备的带组氨酸配基的尼龙66亲和膜在不同离子强度、pH值的盐溶液中及不同温度和流速下对内毒素的去除效果,并对这种亲和介质与内毒素的相互作用机理进行了研究。结果表明:在盐溶液离子强底低于02mol/L、pH值3~8的范围及温度低于50℃、流速小于5mL/min下,可获得最佳去除效果。在选择的最佳条件下能有效去除牛血清白蛋白、氨基酸注射液、人血样等中的内毒素。去除率大部分在90%以上,并能保留住这些样品溶液中绝大多数有效组分,回收率一般都高于80%。
Endotoxin removal under different ionic strength and pH values by nylon 66 affinity membranes with histidine ligands is described. The mechanism of affinity interaction between the membrane and endotoxins is discussed. Effects of temperature and flow rate on endotoxin removal have been investigated. Optimal affinity conditions for endotoxin removal are as follows:ionic strength < 02 mol·L-1, flow rate ≤ 5 mL·min-1, temperature 50 ℃, pH 3~8. Endotoxins can be removed successfully from bovine serum albumin, lysozyme, amino acid injection solution and human blood under optimal affinity conditions.
