摘要
目的研究慢性乙型肝炎(CHB)肝活检组织CCL20的表达与病理分级分期的关系,探讨CCL20在慢性乙肝病毒感染中的作用及意义.方法通过RT—PCR对17例正常肝脏和67例CHB活检组织的CCL20进行半定量(以看家基因GAPDH为对照),结合肝脏组织学积分研究其相关性.结果在HBV慢性感染状态下肝活检组织CCL20的表达水平呈现出个体差异,其CCL20/GAPDH为0.33±0.12,显著低于其在正常肝组织中的平均表达量0.54±0.16(P〈0.01),CCL20/GAPDH在两组之间差异有统计学显著性意义(P〈0.01),CHB患者肝脏中CCL20/GAPDH的表达水平显著降低,且与肝脏的组织学积分相关(r=0.675,P=0.023).结论HBV慢性感染状态下CCL20/GAPDH降低,CCL20的表达与肝脏组织学积分相关.
Objective To investigate the roles and significance of CCL20 expression in liver biopsies of patients with CHB. Methods 67 liver biopsies tissues were used to observe the correlation between HAI of CHB and CCL20 expression. On the basis of established RT-PCR,the expression of the CCL20 in the liver biopsies were semi-quantified (CCL20/GAPDH, GAPDH was considered as housekeeping gene) and at the same time its correlation to HAl were explored. Results Expression of CCL20 in CHB patients showed difference in persons and its CCL20/GAPDH was 0. 33±0. 12,which was lower than that of control groups, which was 0. 54 ± 0. 16 (P〈0.01). That is to say,CCL20 expression in liver biopsies with CHB were decreased than that of in control group(P〈0. 01) and correlation lied in between HAI (Necrosis) and CCL20 expression in liver biopsies of CHB patients(r=0. 675,P=0. 023). Conclusion CCL20 expression was down-regulated in CHB liver biopsies and it showed correlation to HAI of patients,which may be involved in chronic HBV infection.
