Fragments produced by Eco RI digestion of the chromosomal DNA of the glucoamylase producing strain aspergillus niger SG were inserted into the Eco RI site of the vector λ ZAPⅡ,then the recombinant DNA molecules were packaged in vitro to form the genomic library.Through screening the genomic library by hybridization a positive clone was obtained and primarily characterized.The results showed that the cloned 10 kb Eco RI fragment contained the complete A.niger glyceraldehyde 3 phosphate dehydrogenase gene with 2.4 kb in size.
