摘要
目的建立测定那他霉素含量和有关物质的高效液相色谱法。方法本文采用反相高效液相法,用辛烷基硅烷键合硅胶为填充剂;以乙腈-四氢呋喃-乙酸铵溶液(称取3.0g的乙酸铵和1.0g的氯化铵,加水760mL使溶解,摇匀即得)(240∶5∶760)为流动相,流速为2.0mL·min-1,检测波长为303nm,测定了那他霉素的含量。结果那他霉素浓度在0.12~0.28mg·mL-1范围内线性关系很好,r=0.9999;精密度测定RSD=0.11%。结论本方法简便、快速、灵敏、重现性好。
OBJECTIVE To establish a method to assay the content of natamycin and its related substances. METHODS Natamycin was separated and determined on a reverse-phase C8 5mm column with a mobile phase of acetonitrile-tetrahydro-furan-ammonium acetate solution(240 : 5 : 760). The detection wavelength was 303nm. RESULTS The method showed good linearity (r=0.9999) within the range of 0.12-0.28mg·min^-1. The precision testing RSD was 0.11%. CONCLUSION This method is simple and accurate.
