2型肺炎链球菌多糖-破伤风类毒素结合疫苗的制备及免疫原性研究

Studies on preparation and immunogenicity of Streptococcus pneumoniae type 2 polysaccharide-tetanus toxoid conjugate vaccine

摘要 目的制备2型肺炎链球菌多糖(Streptococcus pneumoniae type2 polysaccharide,SPNPS2)-破伤风类毒素(tetanus toxoid,TF)结合疫苗(SPNPS2-TY),并研究其免疫原性。方法采用溴化氰活化SPNPS2,已二酰肼作连接剂,碳二亚胺为耦联剂,将活化的SPNPS2与TT耦联制成SPNPS2-TT.以SPNPS2-TT或单纯SPNPS2免疫Balb/c小鼠,用ELISA检测小鼠血清抗SPNPS2 IgG水平.结果SPNPS2-TT的各项指标均达到质控标准。SPNPS2-TT免疫小鼠产生的特异性抗SPNPS2 IgG水平显著高于单纯SPNPS2免疫小鼠,且SPNPS2-TT诱生了免疫记忆应答。结论采用此项技术制备SPNPS2-TT疫苗是可行的。 Objective To prepare Streptococcus pneumoniae type 2 polysaccharide (SPNPS2)-tetanus toxoid(TT) conjugate vaccine (SPNPS2-TT) and study its immunogenicity. Methods The SPNPS2 was activated by cyanogen bromide. With adipic acid dihydrazide as linking agent,the SPNPS2-TT was prepared by carbodiimide-mediated coupling of SPNPS2 with TT. The Balb/c mice were immunized with SPNPS2-TT or pure SPNPS2, and the auti-SPNPS2 antibodies were detected by ELISA. Results All the qua-lity indexes of SPNPS2-TT met the standards of quality control. The levels of specific IgG antibody elicited in the mice immunized with SPNPS2-TT were significantly higher than those in the mice immunized with pure SPNPS2, and only SPNPS2-TT induced immune memory. Conclusion The SPNPS2-TT is successfully prepared with this technology.
