复方浙贝药物血清影响人脐带间质干细胞增殖与细胞凋亡研究 被引量:3

Research on the effect of Compound Fritillaria thunbergii Miq Particles (CFMP) on the proliferation and inducing apoptosis of human umbilical cord derived mesenchymal stem cells(MSCs)

摘要 目的:观察复方浙贝母药物血清对人脐带间质干细胞(MSCs)增殖抑制和凋亡影响。方法:MTT法测不同剂量组复方浙贝药物血清对人脐带MSCs的抑制率;Annexin-V/PI双染经流式细胞仪(FCM)定量检测复方浙贝药物血清诱导人脐带MSCs细胞早期凋亡率。结果:①高、中、低剂量组复方浙贝药物血清对人脐带MSCs细胞增殖抑制率分别为:3.88%、-3.37%和4.38%;含阿霉素血清组为47.88%。②经FCM双荧光法检测复方浙贝不同浓度药物血清诱导人脐带MSCs细胞早期凋亡百分比不同:空白组为35.19%,低剂量组为1.63%,中剂量组为1.19%,高剂量组为1.70%。结论:复方浙贝药物血清能部分促进人脐带MSCs的增殖(中剂量组),并保护人脐带MSCs的凋亡损伤。 Objective: To investigate the effects of the Compound Fritillaria thunbergii Miq Particles (CFMP) on proliferation and apoptosis of human umbilical cord derived MSCs. Method: MTT was employed to detect the inhibition of the proliferation of MSCs cells at different concentrations of CFMP containing serum which was produced by serum pharmacology method. Cells apoptosis induced by different concentrations of CFMP contained serum were detected by flow cytometry with Annexin-V/PI staining method. Results: ①The inhibition rate of high-group showed 3.88%, middle-group showed 3.37%, low-group showed 4.38%, while DNR-group showed 47.88%. ②Annexin-V/PI staining method showed that the apoptosis rate of MSCs in blank-serum group presented 35.19%, while that of the high-group presented 1.70%, that of the middle-group presented 1.19%, and that of the low-group presented 1.63%. Conclusion: CFMP can partly promote the proliferation of MSCs, and protect the apoptosis of MSCs.
