摘要
目的探索毫米波对细胞质膜通透性及细胞功能的影响。方法以波长7.1mm,功率密度为10mW/cm2毫米波辐照人外周血淋巴细胞15、30、60分钟,并采用钙荧光指示剂Fura-2/Am定量测试法检测淋巴细胞内游离钙浓度和质膜Ca2+Mg2+ATP酶活性变化。结果辐照后淋巴细胞内游离钙浓度明显低于正常(P<0.05);同时细胞膜Ca2+Mg2+ATP酶活性增加(P<0.05);而且辐照后细胞内游离钙浓度降低与质膜上Ca2+Mg2+ATP酶的激活呈负相关。结论一定频率窗和能量窗的毫米波辐照激活细胞膜Ca2+Mg2+ATP酶活性,使细胞膜对钙通透性发生变化,且影响到细胞内Ca2+贮存,造成细胞膜通透性和细胞功能的改变。
Objective To investigate the effect of low power density millimeterwave radiation on cytoplasmic free calcium concentration and the change of plasma membrane Ca2+Mg2+ATPase activity of human peripheral blood lymphocytes.Methods Human peripheral blood lymphocytes,in vitro,randomized into radiated group or control group. Lymphocytes were exposed to 7.1mm millimeterwave for 15,30, 60min, and then the cytoplasmic free calcium concentration in human peripheral blood lymphocytes were measured by fluorescent indicator Fura- 2/Am estimating technique and the change of plasma membrane Ca2+Mg2+ATPase activity in lymphocytes were measured by Lowry's technique.Results The concentration of intracellular plasmic free calcium was lower in radiated group than in control group(P<0.05), and the activity of plasma membrane Ca2+Mg2+ATPase increased significantly. Those changes reduced along with the length of exposing time. It showed that the concentration of intracellular free calcium correlated with the change of plasma membrane Ca2+Mg2+ATPase activity. Conclusion The activity of plasma membrane Ca2+Mg2+ATPase was activated by millimeter wave radiation of certain “radiofrequency window” and “energy window”,and then followed by the change of permeability of plasma membrane, influencing the change of storage of intracellular free calcium and the change of permeability of cell membrane and also cellular function.
