摘要
目的:观察异丙酚对过氧化氢(H_2O_2)介导的内皮细胞损伤的作用。方法:按异丙酚的不同浓度,以脂肪乳剂为对照,用2mmol/L H_2O_导致内皮细胞损伤,将实验分为10组。检测乳酸脱氢酶释放率(LDH)及硫代巴比妥酸反应产物(TBARS)含量,超氧化物岐化酶(SOD)和过氧化氢酶(CAT)活性。并观察细胞形态学改变。结果:临床相关浓度的异丙酚明显降低H_2O_所致的LDH释放,抑制TBARS的产生,保护SOD和CAT活性(P<0.001)。减轻了细胞形态及超微结构的改变。结论:异丙酚对H_2O_2损伤的内皮细胞具有保护作用。
Objective: To observe the effects of propofol on the endothelial cells injured by hydrogen peroxide. Method: Endothelial cells injured by 2mmol/L hydrogen peroxide were divided into ten experimental groups according to the different concentrations of propofol,using intralipid as control. The release of lactate dehydrogenase (LDH), production of thiobarbituric acid reactive substances (TBARS).activities of superoxide dismatase(SOD) and catalase (CAT) were determined as well as morphological change. Result: Propofol at clinical relevant concentrations not only decreased the release of LDH and production of TBARS,but also protected significantly the activities of SOD and CAT (P<0.0001). Conclusion:Propofol can protect vascular endothelial cells against oxidant damage.
