摘要
测定蔷薇红景天提取物中原花青素的含量。采用酸化香草醛比色法,最佳比色条件为:盐酸浓度为8%,香草醛浓度为2%,反应温度为30℃,时间为20min,检测波长500nm。结果表明:该方法的线性范围为10.5μg/mL~420μg/mL,相关系数r=0.9998,平均加标回收率为97.98%,相对标准偏差(RSD)1.24%,检出限为1.32μg/mL。该法操作简便、准确、稳定,适用于原花青素含量的测定。
To establish the method of determining the procyanidins content of Rhodiola rosea extract . Vanillin assay was carried out, the optimum colormetric condition were as follows : the concentration of HC1 was 8 %, vanillin 2.0 %, incubation time for the color formation 20 min and the temperature for color formation 30 ℃, detection wavelength was at 500 nm. There was a good linear relationship between the absorption at 500 nm and concentration in the range of 10.5 μ/mL-420 μ/mL for proeyanidins, and the correlation coefficient was 0.999 8. The average recovery was 97.98 %, RSD was 1.24 % with a detection limit of 1.32 μg/mL. The method is simple, accurate, stabile and can be used for assay of procyanidins.
