摘要
方法:用大鼠肝中血浆硒蛋白PcDNA为探针,对λZPAI大鼠脑cD-NA文库进行筛选,对阳性克隆进行了核酸序列测定。结果:脑中cDNA中蛋白编码区与肝中cDNA的编码区完全一致,但在5utr和3utr存在差异。结论:脑中血浆硒蛋白PcDNA的发现说明脑本身可合成血浆硒蛋白P,这一结果不支持血浆硒蛋白P的硒转运蛋白功能说。
For establishing that selenoprotein P is synthesized in brain,a λZAPII rat brain library was screened with selenoprotein P cDNA of liver.Positive clones were obtained and sequenced.The selenoprotein P cDNA of brain(2041 bp with poly A tail)is shorter than the cDNA of liver(3800 bp).Comparing the sequence of selenoprotein P cDNA coming from brain with the sequence of cDNA of liver,the open reading frame is the same,even though there are some differences in 5utr and 3utr between the two cDNA.The results indicate the selenoprotein P can be expressed in brain too.This results do not support the proposed function of selenoprotein P as selenium transport protein.
