母鼠牛磺酸耗竭与补充对仔代脑发育的影响 被引量:12

摘要 方法:Wistar大鼠随机分成三组:1.牛磺酸耗竭组(GES组),母鼠孕15日至哺乳第17日向饮水中加入GES(浓度为1%),并给予牛磺酸缺乏膳(饲料牛磺酸含量为0);2.对照组(Con组)饲料牛磺酸含量为0.06%,饮水中不含添加物;3.牛磺酸补充组(TS组),饲料牛磺酸含量为0.6%,饮水中亦不含添加物。对仔代出生及断乳时的脑重、脑牛磺酸与游离氨基酸含量、脑微量元素Zn、Cu、Fe含量及学习记忆能力进行测定。牛磺酸测定采用离子交换层析法;氨基酸测定采用HPLC法;微量元素Zn、Cu、Fe测定采用原子吸收火焰法;乳鼠学习记忆能力测定采用Y迷宫进行。结果:GES组仔鼠出生及断乳时的脑重、脑牛磺酸与微量元素Zn含量均低于对照组;在Y迷宫中的学习记忆能力低于母鼠牛磺酸补充组。母鼠牛磺酸补充对仔鼠出生时的脑重、脑牛磺酸含量及与微量元素Zn含量均无影响;但至断乳时,仔鼠大脑牛磺酸含量、微量元素Zn、Fe含量高于正常对照组。 In order to study the effect of taurine level in pregnant rats on the development of offsprings brain,a domestic prior study using guanidinoethyl sulfonate(GES) as an inhibitor of taurine in the pregnant rats was conducted.Pregnant Wistar rats were randomly divided into three groups:(1)GES groups,rats were fed with a taurinefree diet,and 1% GES was added to drinking water during the period of the 15th day of pregnancy to the 17th day of lactation.(2)Control group,rats were fed with normal fodder containing 0.06% taurine,no GES was added to the drinking water.(3)Taurine supplemented group,fodder contained 06% taurine,and no GES was used.Brain weight and taurine,free amino acids and trace elements of brain were measured in the offspring at postnatal day 2 and day 21(weaning).The learning and memory ability was tested by Y maze.The results showed that brain weight,brain taurine and zinc content of the pups in GES group were lower than those of control at postnatal day 2 and day 21;and learning and memory ability was lower than those of the pups in taurine supplemented group.
