摘要
目的研究在大鼠肾移植中骨髓干细胞移植对免疫耐受的诱导作用及机制。方法40只Wistar大鼠(雌雄各半)随机分为实验组和对照组,进行肾移植手术,术前经门静脉分别注射骨髓细胞及生理盐水,30天后观察肉眼和光镜下排斥反应及检测嵌合体的形成。结果实验组肉眼和光镜下排斥反应均较轻微,嵌合体检测为阳性。结论应用他克莫司并注射供体骨髓细胞,可诱导一定的免疫耐受。移植前输注供体骨髓细胞而形成的嵌合体与免疫耐受有关。
Objective To study the protective effectiveness of immunogical tolerrance of kidney transplantation in rats by bonemarrow transplantation. Methods Forty Wistar rats were randomly assigned into the control group (n = 20) and experimental group (n = 20). The female animals were injected bonemarrow cell and normal salty in portal vein. After 30 days detection ehimerism of blood sample and kidneys tissue regular - dye were studied. Results In experimental group positive rates of chimerism were significantly different and rejection reaction was significantly lower than that of control group. Conclusion Bonemarrow cell injection could induce certain extent of immunogical tolerrance. Donor bone marrow cells injection before the transplantation may work through the mechanism of establishment of chimerism.
