GCS和MDR-1基因在CML中的表达及意义

Expression and significance of GCS and MDR-1 gene in patients with chronic myeloid leukemia

摘要 目的观察慢性粒细胞白血病(CML)患者葡萄糖神经酰胺合酶(GCS)和多药耐药1(MDR-1)基因的表达变化及其意义。方法建立实时荧光逆转录聚合酶链反应(RT-PCR)检测GCS、MDR-1和磷酸甘油醛脱氢酶(GAPDH)基因的方法,检测CML初发组25例和急变组25例患者外周血GCS、MDR-1基因表达水平,以GAPDH为内参基因,两组基因表达水平的差异采用△△CT相对定量法。结果外周血GCS水平CML急变组较初发组显著增高(4.84倍,P<0.01),MDR-1表达CML急变组9例阳性(36%),CML初发组均为阴性。结论GCS和MDR-1基因高表达在CML急变患者的多药耐药中起着重要作用,其水平检测可为CML急变预后判断的新指标。 Objective To study the expression and significance of glucosylceramide synthase (GCS) and multiple drug resistance 1 (MDR-1 ) gene in the patients with primary or blast crisis chronic myelogenous leukemia (CML). Methods GCS, MDR-1 gene and glyceraldehyde phosphate dehydrogenase(GAPDH) in 25 PBMC samples with primary CML(group A) and in 25 cases with blast crisis CML(group B) were detected by real-time RT-PCR. The difference of gene expression between the two groups was analyzed by AACt relative quantity means, in which GAPDH gene was used as the control. Results The expression of C, CS gene in group B was 4.84 times as much as that in group A (P〈0. 01). The positive expression rate of MDR-1 gene 36%(9/25) in group B, but MDR-1 gene was expressed negitively in group A. Conclusion The overexpressions of GCS and MDR-1 genes play an important role in MDR during blast crisis progression of CML, which could be taken as a new marker in predicting the prognosis in blast crisis CML.
