蜕皮甾酮对大鼠同种异体心脏移植的影响 被引量:3

Effect of ecdysterone on heteroptopic heart transplantation in rats

摘要 目的探讨蜕皮甾酮(EDS)对大鼠同种异体心脏移植的保护作用。方法随机取供体、受体各20只SD大鼠建立腹腔心脏移植动物模型,受体术后EDS组(10只,每日腹腔注射EDS20 mg/kg),对照组(10只,每日腹腔注射生理盐水);另10只为仅开关腹腔假手术组。术后第7天,取移植心脏病理学观察细胞形态及透射电镜观察超微结构;移植受鼠和假手术组,采血测血清心肌酶谱、氧化代谢产物。TUNEL技术和ABC法免疫组化染色检测细胞凋亡指数(AI)及相关基因了解细胞凋亡情况。结果病理切片和透射电镜观察均提示EDS组心肌细胞损害轻于对照组。与对照组相比,EDS组血清各心肌酶含量明显减少(P<0.01),氧化代谢产物明显减少。对照组与EDS组相比AI上升。EDS组凋亡相关基因Bcl-2表达增强,Bax表达减弱。结论EDS对移植心脏有多种保护作用。 Objective To investigate the protective effects of ecdysterone(EDS) on the allograft heart transplant model of rats. Methods The healthy SD rats were randomly assighned to the donors and recipients. The hearts were placed heterotopically into allogenic recipients and harvested on the 7^th day. All animals were divided into three groups of sham operation group(only openning and cloing the abdomen,n= 10), EDS group (injecting intraperitoneally with EDS 20 mg·kg^-1·day^-1, n= 10), control group (injecting intraperitoneally with normal saline, n = 10). The pathological changes of myocardiacal tissues were analyzed by light microscopy and transmission electron microscopy and the levels of myocardial enzymes, SOD, ET-1, NO, MDA in serum were measured. Tissue samples were underwent the detection of apoptotic cell death by in situ terminal deoxyribonucleotide transferasemediated dUTP nick end labeling for apoptotic index (AI) and also using immunohistochemistry method to study the expression of Bcl-2 and Bax. Results Pathological examination and transmission electron microscope observation showed a greater myocardium damage in control group. EDS group showed a decrease in the amount of myocardial enzymes, MDA, ET-1 and an increase in the levels of SOD, NO compared to those in control group. The AI of control group became higher than that of EDS group, meanwhile Bcl-2 expression was increased and Bax decreased. Conclusion EDS has some protective effects on heteroptopic heart transplantation.
