摘要 本文对脑组织切片培养技术进行了详细的介绍,包括仪器设备、培养液的配制、脑组织切片制备及培养过程等。利用此技术,本文作者对大鼠脑海马结构及其苔藓纤维进行了组织学、组织化学和生物素标记研究。结果显示,在体外培养条件下海马结构的器官结构特征被保持,苔藓纤维发育良好,并与相应的靶细胞建立了突触联系,与体内研究结果一致。这表明脑组织切片培养技术是在体外研究神经元发育和神经纤维联系的理想方法。 In this paper the detailed procedures for slice culture technique of brain tissue were introduced, including the equipments required, reagents, preparation of brain tissue slice and incubation. With this technique the rat hippocampal formation and its mossy fiber were studied for histology, histochemistry and biocytin tracing. The results showed that under culture condition in vitro the organotypic organization of hippocampal formation was retained, the mossy fibers developed wel1 and formed synaptic connections with their target cells. These results are in agreement with the reP0rts in vivo. This illustrates tFat the slice culture technique of brain tissue is an ideal tool for the studies of neuronal development and nerve fiber connections in vitro.
