DNA Analysis of an Ancient Corpse of Ming Dynasty Found in Yunnan

目的对距今475a的古代遗骸进行DNA分析,为考古人类学DNA分析完善资料和积累经验.方法以经典的酚/氯仿法和chelex-100法提取DNA,以Qiaquick PCR Purification Kit对DNA进行纯化,以Powerplex 16 Kit进行PCR复合扩增并对结果进行检测分型.结果STR位点图谱清晰.结论复合扩增法可用于古尸DNA分析.

Objective To provide beneficial materials and experience for anthropology＇ s DNA analysis by analyzing an ancient corpse that was 475 years ago. Methods DNA was extracted by methods of phenol-chloroform and chelex-100 and purified by Qiaquick PCR Purification kit. The STR loci were amplified with Promega Powerplex16 system. Result Distinct STR locus map was clearly obtained. Conclusion Ancient DNA can be well analyzed by this method.
