摘要
目的建立核定位信号的实验教学模型.方法构建pdx-1基因和EGFP基因融合表达的表达载体,脂质体转染法将载体转入培养的HeLa细胞,荧光显微镜下观察.结果转染的HeLa细胞仅在细胞核发荧光,证明pdx-1蛋白的核定位信号将融合蛋白EGFP-pdx-1引导到细胞核.结论用转录因子pdx-1和增强型绿色荧光蛋白EGFP建立核定位信号的实验教学模型是可行的.
Objective To establish an experimental teaching model of nuclear localization signal. Methods A fusion gene expression vector with EGFP and pdx-1 was constructed, HeLa cells were transfected with the expression vector using LIPOFECTAMINETM Reagent (Gibco), transfected cells were observed under fluo- rescence microscope. Results Fluorescence was only detected in the nucleus of the transfected HeLa cells, this indicated that nuclear localization signal in the pdx-1 induced the EGFP-pdx-1 fusion protein to enter into the nucleus. Conclusion It is feasible to establish the experimental teaching model of nuclear localization signal with pdx-1 and EGFP.
