摘要
为探讨MAC亚溶破效应在移植肾慢性排斥反应发病机制中的作用和CD59的保护作用,建立了肾小球血管内皮细胞培养方法,并用酵母多糖(Z)激活血清(NHS)补体,诱生MAC损伤肾小球血管内皮细胞,以上清中LDH释放量及脱落细胞活力确定NHS的亚溶破损伤剂量。结果显示:用肾小球外植法培养肾小球血管内皮细胞较易成功,当NHS为0.05ml(5%)时,上清中的LDH和脱落细胞活力,与正常组比较相差显著(P<0.05)。
To study the role of the sublytic effect of MAC in the mechanism of chromic allograft rejection and pretection of CD59 on it,the culture of glomerlar endothelial cells was modinfied and the sublytic damage of MAC on the cells was determed with measuriug the changes of LDH in supernatant of the cells damaged by actived complement.The result shown that the culture of glomerualar endtholial cells was easily successful with the transplant method of isolated glomerali and the sublytic damage on the cells was resulted from 5% mixed NHS.
