替米沙坦对2型糖尿病大鼠心肌保护作用及机制 被引量:1

Protective Effect of Telmisartan on Cardiomyopathy in Diabetic Rats and Its Mechanism

摘要 目的:探讨替米沙坦对2型糖尿病大鼠心肌保护作用及其细胞因子机制。方法:尾静脉注射链脲佐菌素(STZ)复制糖尿病大鼠(DM)模型。正常对照10只(C组),糖尿病模型12只不喂替米沙坦(D组),12只喂替米沙坦(T组)。12周末检测心功能及电镜观察心肌超微结构,western blot方法检测心肌HGF含量。结果:三组比较,LVSP和+dp/dtmax均无差异。与C组比较,D组及T组LVEDP升高(P<0.01),-dp/dtmax降低(P<0.05),HGF升高(P<0.01);与D组比较,T组LVEDP降低(P<0.01),-dp/dtmax升高(P<0.05),HGF升高(P<0.01)。结论:替米沙坦保护糖尿病大鼠心肌,可能与升高心肌HGF有关。 Objective:To investigate the protective effect of telmisartan on cardiomyopatby in diabetic rats and its mechanism. Method:The models of diabetic rats were made by a tail-vein injection of streptozotocin (STZ). Ten rats were the normal control rats. Tewelve diabetic rats were not feeded with telmisartan and Tewelve diabetic rats were feeded with telmisartan. At the time of 12 weeks feeded with telmisartan,the cardiac function were measured;ultrastructure of eardiomyocytes was observed under electron microscope; expression of HGF in eardiomyopathy were measured with western blot method. Results:LVSP and +dp/dtmax had no differents among the three groups. LVEDP were much higher (P〈0. 01),-dp/dtmax were declined(P〈0.05),HGF were evated (P〈0.01)in D group and Tgroup,compared wiht C group. LVEDP was declined(P〈0.01), dp/dtmax was elevated(P〈0. 05),HGF of T group was evated(P〈0. 01 ) in T group compared wiht D group. Conclution :Telmisartan can protect cardiomyopathy of diabetic rats, which may be related to the increation of HGF in myocardition.
