摘要
采用改良WPM为基本培养基,研究蓝莓的组培育苗技术。结果表明:以当年生绿枝茎段为外植体可有效获得无菌苗;蓝莓茎段的离体增殖率受到培养基中细胞分裂素的调控,ZT 1-2mg/Li和2ip 10~20mg/L的效果较好,初代培养增殖率3-6倍。继代培养时茎段和叶片也可分别再生无菌苗6-8周继代1次,增殖率可达10倍以上。蓝莓试管苗瓶内生根率不高,0.02~0.2mg/L IBA可诱导蓝莓生根,正常生根率30%~50%。
The modified WPM was taken as the basic medium to study blueberry tissue culture technology. The results showed that the virus-free plantlets could be obtained after taking the green shoots as the explants. The shoots were well proliferated on WPM medium supplemented with ZT 1.0-2.0 mg/L or 2ip 10-20 mg/L, and the multiplication rate of initial culture was 3-6 times. The shoots and leaves asepsis plantlets could also produce virus-free plantlets on the same medium and the multiplication rate of every subculture was over 10 times during 6-8 weeks. The rooting rate in the bottle was not high, which usually was 30%-50%, and 1/2 WPM with the IBA 0.02-0.2 mg/L could be used as rooting medium.
