摘要
【目的】观察地塞米松对肺腺癌细胞生长及与化疗作用的影响并探讨其与凋亡相关基因突变型p53及耐药P糖蛋白(P-glycoprotein,P-gp)表达相关性。【方法】将不同浓度的地塞米松单独或与长春新碱(VCR)联合作用于肺腺癌细胞株A549。四唑盐(MTT)试验测定药物对肺腺癌细胞A549的抑制率,筛选出药物的IC50浓度。免疫组化法测定IC50浓度各药物应用对p53、P糖蛋白(P-gp)表达的影响,比较二者的相关性。【结果】不同浓度的地塞米松对肿瘤的生长及化疗有影响,且上述药物与IC50浓度的VCR联合应用时能增加VCR的抑制率,用药后各组p53、P-gp表达均减少(P<0.01),且二者表达呈正相关。【结论】地塞米松对A549细胞的生长及化疗有影响,其机制可能与凋亡相关基因p53及P-gp介导的肺癌耐药机制有关。
[Objective] To observe the impact of dexasone on the growth of pulmonary adenocarcinoma cell A549. Furthermore to study the influence is dued to affecting the expression of the two protein p53 and P-Glycoprotein (P-gp). [Methods] To place different concentration of dexasone on A549 cell alone or combinated with xincristine (VCR). MTF assay (mononuclear cell direct cytoxicity assay) : to estimate the antirate of those medicine on A549 cell, screening the concentration of IC50 in those medicine. Immunohistology chemistry Technique (IHC) To determinate the influence of those medicine, which with xincristine (VCR) of IC50, on the expression of p53 and P-gp , moreover compare the correlation of the two protein. [Results] dexasone all have effect on the growth of A549 cell. IC50 concentration on A549 cell make significantly change of the expression of p53 and P-gp, all is decreased/decereaced (P 〈 0.01 ) and the expression of p53 is positively correlated. [Conclusions] The result show that dexasome can have an effect on the growth of tumor in vitro, which make clear that p53 and P-gp may be involved in the pathways of multidrug resistance.
