摘要
目的:电子克隆获得萝卜干细胞决定基因WUS的编码序列。方法:利用已知的拟南芥WUS基因序列,采用电子克隆的方法获得了萝卜WUS基因cDNA的序列。并对萝卜WUS的氨基酸序列组成,物理和化学性质,二级的特点以及其与其他植物WUS的进化关系进行了研究。结果:萝卜WUS基因cDNA全长为1 298 bp,包含一个完整的开放读码框(936 bp),编码312个氨基酸。其分子量为35 465.7 Da,等电点为6.99。其与拟南芥WUS的同源性为74.0%。在进化关系上,萝卜与油菜和拟南芥WUS亲缘关系较近。与豆科植物亲缘关系远。结论:初步获得了萝卜WUS基因的编码序列,为进一步研究其功能打下基础。
Objective:Obtain the sequence coding WUS gene,stem cell organizer gene,in Raphanus sativus.Method:With WUS gene in Arabidopsis thaliana,the cDNA of WUS gene in Raphanus sativus was in silico cloned.Some characters of the WUS,such as the composition of amino acid sequences,physical and chemical properties,secondary and the evolutionary relationship were studied.Result:The length of cloned WUS cDNA was 1 298 bp and it contained a complete ORF(936 bp),encoded 312 amino acids.The calculated molecular weight of it was 35 465.7 Da,theoretical pI of 6.99.It has homology of 74.0% with the Arabidopsis wus.On the evolutionary relationship,Raphanus sativus wus was close to Brassica napus and Arabidopsis.And phylogenetic relationship was far to Leguminosae plants.Conclusion:The WUS gene coding sequence of Raphanus sativus was cloned preliminary,and laid the foundation for further study its function.
