漏斗胸大鼠肺泡表面活性物质中磷脂的测定及其意义 被引量:2

Investigation and its significance of phospholipid in alveolar surfactant with acquired pectus excavatum in rats

摘要 目的探讨获得性漏斗胸形成过程中肺泡表面活性物质中磷脂含量的变化。方法取同批不同窝4周龄SD大鼠54只,随机分成实验组和对照组,其中实验组30只,通过胸骨旁切断下位三对肋软骨复制漏斗胸模型,对照组24只,不做任何干预。分别于术后2周、4周和8周实验组各处死10只和对照组各处死8只取大鼠肺脏,收集并提取肺泡表面活性物质中的磷脂成分,经Trinder反应后通过比色法测定磷脂含量,对结果进行单因素方差分析和组间t检验。结果实验组动物术后均出现前胸壁漏斗状畸形。各时间点测得肺泡表面物质中磷脂含量在组间比较,术后2周实验组的测得值高于对照组,而术后4周和8周却低于对照组,三个时间点的磷脂测得值呈递减趋势,差异有统计学意义(P<0.01),而对照组各时间点测得值的差异无统计学意义(P>0.05)。结论漏斗胸造模术后大鼠肺泡表面活性物质中磷脂含量呈先高后低并逐渐递减的双相变化,提示在获得性漏斗胸形成过程中肺泡的顺应性发生了改变。 Objective To explore the variation of phospholipid(PL)in alveolar surfactant in the process of pectus excavatum(PE)occurred on young rats.Methods 54 Sprague Dawley(SD)rats at 4 weeks old were divided into two groups randomly.The lower three couples of costal cartilage were cutted off side by sternum for model of PE in 30 rats as group one and no intervene in another 24 rats as a control.The lungs of 10 rats in group one and 8 rats in control were harvested at 2nd,4th and 8th week after operation,PL in alveolar surfactant were collected from flushing fluid of lungs and determined through Trinder reaction and colorimetric method.The data were analyzed statistically with One-way ANOVA and t-Test.Results PE deformation occurred in all animal of group one after operation.As a result,there was significance statistical differentiation compared the value of PL between two groups and it was higher at 2nd week and lower at 4th and 8th week in group one than that in control(P<0.01).The PL value varied with tendency of decreasing progressively in group one but the PL value variation with no significance statistical differentiation compared within control group among three time point(P>0.05).Conclusions It is occurred that dual phase change of PL value in alveolar surfactant with increasing at first and then reducing progressively and which suggested that the alveolar compliance changes in the process of acquired pectus formation in young rats.
