缬沙坦对自发性高血压大鼠心脏、胸主动脉血管紧张素转换酶2表达的影响 被引量:3

The effect of valsartan on the expression of angiotensin converting enzyme 2 in the heart and aorta of spontaneously hypertensive rats

摘要 目的探讨血管紧张素转化酶2(ACE2)在自发性高血压大鼠(SHR)心脏及胸主动脉中的表达以及缬沙坦对其的影响。方法 24只SHR随机分为SHR组(n=12)、缬沙坦组(n=12),12只血压正常的Wistar大鼠作为正常对照组,缬沙坦组每只大鼠给予缬沙坦30mg.kg-1.d-1灌胃,SHR组及正常对照组给予等量生理盐水灌胃。10周后,应用光镜观察心肌的病理变化,测定主动脉中膜厚度与血管腔内径之比。免疫组化法检测心肌、主动脉中ACE2的表达。结果与正常对照组比较,SHR组心肌、主动脉中ACE2表达显著降低(P<0.05);与SHR组比较,缬沙坦组ACE2表达显著增高(P<0.05),缬沙坦组血压、心脏重量指数较SHR组均明显下降(P<0.05)。结论自发性高血压大鼠心肌、主动脉中ACE2表达降低,缬沙坦能够上调SHR心肌、主动脉中ACE2的表达,该作用可能为血管紧张素Ⅱ1型受体拮抗剂抑制高血压病所导致的心血管重构的新机制。 Objective To investigate the regulation effect of valsartan on the expression of ACE2 in the heart and aorta of spontaneously hypertensive rats(SHR).Methods Twenty-four 12 week-old male SHR were randomly divided to SHR group(n = 12) and valsartan group(n = 12) ;Twelve 12 week-old male Wistar rats were served as control group.Valsartan group was given daily dose of 30 mg /kg valsartan for 10 weeks and the other two groups were given equal saline.After 10 weeks,the pathological changes of the heart were observed,the rate of intima media thickness and lumen diameter of the aorta were determined.The expression level of ACE2 in heart and aorta was measured by immunohistochemistry.Results The ACE2 protein expression was significantly down regulated in the heart and aorta of SHR group compared with normal group(P < 0.05).After 10 weeks treatment with valsartan,the attenuated ACE2 expression in valsartan group was markedly enhanced compared with SHR group(P < 0.05).The systolic pressure and heart quality index were decreased as well.Conclusions The expression level of ACE2 in SHR is reduced and valsartan could increase the expression of ACE2.It is the new mechanism that angiotensin receptor blocker inhibit the cardiovascular reconstruction resulting from hypertension.
